Mechanism of multidrug resistance of "Superbacteria" clarified

Recently, XJTU researchers conducted a study on the regulatory mechanism of multi-drug resistance of "super bacteria". By applying integrated approaches to structural biology, researchers revealed that the response regulatory factor AdeR can control the expression of the AdeABC efflux pump by recognizing a DNA sequence in the intercistronic region between AdeR and AdeABC. Researchers also introduced a 1.4 (ngstrom, 1 ngstrom = 10-10m) AdeR regulatory domain, a 1.6 complex of AdeR regulatory domain bound with magnesium ions, and a 2.75 complex of AdeR DNA-binding-domain with its targeted 25bp intercistronic DNA in crystal structure. In addition, small angle X-ray scattering was applied to verify the assembling properties of AdeR under physiological conditions, thus systematically explaining the direct regulation of the molecular regulating mechanisms of the response regulatory factor AdeR to control the AdeABC efflux pump. This provides a reference for finding new therapies of multi-drug resistant AcinetobacteRbaumannii and developing new antibiotics.

The above findings were published in the Nucleic Acids Research (impact factor 10.162) with the title of "Mechanistic Insight into How Multidrug Resistant Acinetobacter Baumannii Response Regulator AdeR Recognizes an Intercistronic Region".
